Overview

Deflazacort is a glucocorticoid used as an anti-inflammatory and immunosuppressant. Deflazacort is an inactive prodrug which is metabolized rapidly to the active drug 21-desacetyl deflazacort. Its potency is around 70 to 90% that of prednisone. Deflazacort, the active ingredient of ATMOCORT tablets is a corticosteroid. Corticosteroids reduce inflammation and suppress allergic reactions and immune system activity.

Indications

ATMOCORT tablets are used to treat a variety of inflammatory conditions including asthma, arthritis, allergic disorders and certain disorders of the skin, kidney, heart, digestive system, eyes and blood as well as some tumour-producing diseases. They are also used in transplant operations.
ATMOCORT can be used to:

  • Treat inflammation including asthma, arthritis and allergies
  • Treat problems with your skin, kidney, heart, digestive system, eyes or blood
  • Treat problems where your body has growths (tumors)
  • Suppress the immune system in transplant operations.

Use in Pregnancy and Lactation

Deflazacort does cross the placenta. However, when administered for prolonged periods or repeatedly during pregnancy, corticosteroids may increase the risk of intra-uterine growth retardation. As with all drugs, corticosteroids should only be prescribed when the benefits to the mother and child outweigh the risks.

Warning

The following clinical conditions require special caution and frequent patient monitoring is necessary:
Cardiac disease or congestive heart failure (except in the presence of active rheumatic carditis), hypertension, thromboembolic disorders. Glucocorticoids can cause salt and water retention and increased excretion of potassium. Dietary salt restriction and potassium supplementation may be necessary.
Gastritis or oesophagitis, diverticulitis, ulcerative colitis if there is probability of impending perforation, abscess or pyogenic infections, fresh intestinal anastomosis, active or latent peptic ulcer.
Diabetes mellitus or a family history, osteoporosis, myasthenia gravis, renal insuffciency.
Emotional instability or psychotic tendency, epilepsy.
Previous corticosteroid-induced myopathy.
Liver failure.
Hypothyroidism and cirrhosis, which may increase glucocorticoid effect.
Ocular herpes simplex because of possible corneal perforation.

Contraindications

Rheumatoid Arthritis & Juvenile Chronic Arthritis, Asthma and other airway diseases, Pemphigus, Uveitis, Nephrotic Syndrome, immunne suppression in transplantation.
Hypersensitivity.

Side Effects

GI disturbances, musculoskeletal, endocrine, neuropsychiatric, ophthalmic, fluid and electrolyte disturbances; susceptible to infection, impaired healing, hypersensitivity, skin atrophy, striae, telangiectasia,acne, myocardial rupture following recent MI, thromboembolism.

Dosage

This is Prefered Dosage:
Adults: 6mg.
Hepatic Impairment: Dose should be carefully monitored and adjusted to the minimum effective dose.
Elderly: No special precautions other than those usually adopted in patients receiving glucocorticoid therapy are necessary.
Children: Doses of deflazacort usually lie in the range 0.25 -1.5mg/kg/day.

  • Painkillers such as aspirin
  • Aminoglutethimide used for some types of cancer
  • Ketoconazole used to treat infections
  • Medicines for thinning your blood (such as warfarin)
  • Medicines for diabetes
  • Medicines for epilepsy such as phenobarbitone, primidone, phenytoin, carbamazepine, acetazolamide
  • Medicines which contain oestrogens including oral contraceptives
  • Medicines for tuberculosis (TB) such as rifampicin or rifabutin
  • Medicines for high blood pressure

For Patient

Do not take this medicine and tell your doctor if:
  • You are allergic (hypersensitive) to deflazacort or any of the other ingredients in these tablets Signs of an allergic reaction include: a rash, swallowing or breathing problems, swelling of your lips, face, throat, or tongue.
  • You have an infection that affects your whole body (systemic infection), which is not already being treated.
  • You are having or have recently had any vaccinations with live viruses.
