Overview
This medication is used to treat a wide variety of bacterial infections. This medication is known as a cephalosporin antibiotic. It works by stopping the growth of bacteria. Cefpodoxime and Ofloxacin is broad-spectrum antibiotic which are used to treat a variety of infections caused by bacteria like pneumonia, sinusitis, cystitis and ear infections. Antibiotics such as cefpodoxime will not work for colds, flu, or other viral infections.
Indications
- Bacterial Infections
- Urinary Tract Infections
- Eye and Ear infections
- Acute bacterial exacerbation of chronic bronchitis
Contraindications
Cefpodoxime Proxetil:
Hypersensitivity reactions to cefpodoxime, other cephalosporins, penicillins, or other drugs.
If cefpodoxime is to be administered to penicillin sensitive patients, caution should be exercised because cross hypersensitivity among beta-lactam antibiotics has been clearly documented and may occur in up to 10% of patients with a history of penicillin allergy. .
Ofloxacin:
Ofloxacin is contraindicated for the treatment of certain sexually transmitted diseases by some experts due to bacterial resistance.
Caution in patients with liver disease. The excretion of ofloxacin may be reduced in patients with severe liver function disorders (e.g., cirrhosis with or without ascites).
Ofloxacin is also considered to be contraindicated within the pediatric population, pregnancy, nursing mothers, and patients with psychiatric illnesses and in patients with epilepsy or other seizure disorders.
For Professionals
Pharmacodynamics Properties of Cefpodoxime:
Cefpodoxime is an oral third generation cephalosporin antibiotic. It is active against most Gram positive and Gram negative bacteria Cefpodoxime is active against a wide spectrum of Gram-positive and Gram-negative bacteria. Cefpodoxime is stable in the presence of beta-lactamase enzymes. As a result, many organisms resistant to penicillins and cephalosporins, due to their production of beta-lactamase, may be susceptible to cefpodoxime. Cefpodoxime is inactivated by certain extended spectrum beta-lactamases. The bactericidal activity of cefpodoxime results from its inhibition of cell wall synthesis. The active metabolite of cefpodoxime binds preferentially to penicillin binding protein 3, which inhibits production of peptidoglycan, the primary constituent of bacterial cell walls.
Pharmacokinetics Properties of Cefpodoxime:
Cefpodoxime proxetil is a prodrug that is absorbed from the gastrointestinal tract and de-esterified to its active metabolite, cefpodoxime and protein binding is 22 to 33% in serum and from 21 to 29% in plasma. About 29% to 33% is excreted unchanged in the urine in 12 hours.
Pharmacodynamics Properties of Ofloxacin:
Ofloxacin is a quinolone/fluoroquinolone antibiotic. Ofloxacin is bactericidal and its mode of action depends on blocking of bacterial DNA replication by binding itself to an enzyme called DNA gyrase, which allows the untwisting required to replicate one DNA double helix into two. Notably the drug has 100 times higher affinity for bacterial DNA gyrase than for mammalian. Ofloxacin is a broad-spectrum antibiotic that is active against both Gram-positive and Gram-negative bacteria.
Ofloxacin acts on DNA gyrase and toposiomerase IV, enzymes which, like human topoisomerase, prevents the excessive supercoiling of DNA during replication or transcription. By inhibiting their function, the drug thereby inhibits normal cell division.
Pharmacokinetics Properties of Ofloxacin:
Absorption:Bioavailability of ofloxacin is the tablet formulation is approximately 98%
Protein Binding:32%
Metabolism:Hepatic
Excretion: Elimination is mainly by renal excretion.
